What is retatrutide? The research in brief
Retatrutide (LY3437943) is an investigational once-weekly injection developed by Eli Lilly. It is a single peptide that activates three hormone receptors — GIP, GLP-1 and glucagon — which is why it is often called a "triple agonist." Approved medicines such as semaglutide act on GLP-1 alone, and tirzepatide on GLP-1 and GIP.
- Phase 2 (2023): in a 48-week trial of 338 adults, the 12 mg dose reduced body weight by an average of 24.2%, versus 2.1% on placebo (Jastreboff et al., New England Journal of Medicine).
- Phase 3 TRIUMPH-1 (2026): 28.3% average weight loss at 80 weeks on 12 mg (efficacy estimand), or 25.0% counting every randomized participant.
- Side effects in trials: mainly nausea, diarrhea and vomiting; up to 11–18% of participants on the highest dose stopped treatment because of adverse events.
- Approval status: not approved by the FDA. Lilly plans to submit it for U.S. approval in the first quarter of 2027.
The FDA states that retatrutide cannot be compounded and has issued warning letters to sellers that label it "for research use only" while marketing it for human use; it urges consumers not to buy these products (FDA: concerns with unapproved GLP-1 drugs).
